“Lethal Weapon” star Danny Glover announced that he’s been diagnosed with Alzheimer’s disease.
The 79-year-old actor discussed his diagnosis alongside family members in interviews with multiple outlets. 
“I’m sure as it advances, things are going to be different and changing,” Glover said in an <a href="https://www.today.com/health/celebrity-health/danny-glover-alzheimers-exclusive-today-rcna352405">interview</a> with Today that aired on Wednesday. 
The “Field of Dreams” star revealed that he was diagnosed in 2023. He’s now working with the Alzheimer’s Association to spread awareness about the progressive, fatal neurodegenerative disease. 
“I&#8217;m still not accepting in my mind all parts of it,” the actor told People in a separate <a href="https://people.com/danny-glover-opens-up-alzheimers-diagnosis-79-life-continues-exclusive-12010257">interview</a>. “There are the moments that you keep remembering that validate the fact that you can remember stuff. And there are moments I&#8217;ll never forget.”
Glover rose to prominence with roles in “Places in the Heart” (1984), “The Color Purple” (1985), and the Lethal Weapon franchise. He’s been involved in most genres, from family-friendly films to action to drama. 
“I think it’s really important for him to have control of his own narrative, of his own life story,” Glover’s daughter, Mandisa, told NBC News journalist Lester Holt during the interview. “That’s really important. And the time is now. What better time but now for him to speak for himself?&#8221;
“It’s important because people ask questions sometimes, and I don’t want to be a dishonest person and say, ‘Oh, yeah, everything is all right. It’s all great,’” she said.
Mandisa said she believes the disease is likely “depressing” to live with. “It&#8217;s a change in the core of who you think you are or don&#8217;t think you are,” she told the outlet, saying it’s “very hard” for the family to deal with. Glover’s younger brother Marty said, “We just want him to live his best life like he made us live ours.”
Despite the diagnosis, Glover expressed optimism about the future. 
“I don&#8217;t feel like it&#8217;s the end of my life,” Glover told People. “&#8230;I still have my daughter, I have friends,” he added. “I want to just say, your life continues.”
Outside of acting, Glover has long been active in left-wing political causes, endorsing presidential campaigns of Sen. Bernie Sanders (I-VT) in both 2016 and 2020 and participating in numerous protests.

“The Notebook” star Gena Rowlands’ son Nick Cassavetes shared heartbreaking news that his mother is in “full dementia” and has been diagnosed with <a href="https://www.dailywire.com/news/chris-hemsworth-said-hes-taking-these-steps-to-combat-increased-alzheimers-risk" target="_blank" rel="noopener">Alzheimer’s disease</a>.
During an interview with Cassavetes, in honor of the romantic film’s 20th anniversary, the director and son of the 94-year-old actress reflected on the character his mom played in his 2004 film — the older version of Allie, a woman with dementia — while Rachel McAdams played the younger one, <a href="https://ew.com/the-notebook-star-gena-rowlands-has-alzheimers-8668642">Entertainment Weekly</a> reported.
“I got my mom to play older Allie, and we spent a lot of time talking about Alzheimer’s and wanting to be authentic with it, and now, for the last five years, she’s had Alzheimer’s,” Cassavetes said.
“She’s in full dementia,” he added. “And it’s so crazy — we lived it, she acted it, and now it’s on us.”
Speaking on the film’s anniversary, Cassavetes also said that he’s proud of the film they made together.
It &#8220;holds up pretty good after 20 years,” the director said.  &#8220;It&#8217;s always a shock to hear that as much time has gone by as it has, but it makes sense.”
“I&#8217;m just happy that it exists,&#8221; he added.  &#8220;It seems to have worked and I&#8217;m very proud of it.&#8221;
After the movie came out in 2004, Rowlands spoke to <a href="https://www.oprah.com/spirit/gena-rowlands-aha-moment">O magazine</a> about relating to the character as her own mother, actress Lady Rowlands also had been diagnosed with the disease.

“This last one — ‘The Notebook,’ based on the novel by Nicholas Sparks — was particularly hard because I play a character who has Alzheimer’s,” Rowlands said at the time.
“I went through that with my mother, and if Nick hadn’t directed the film, I don’t think I would have gone for it — it’s just too hard,” she added. “It was a tough but wonderful movie.”

<a href="https://www.dailywire.com/topic/chris-hemsworth">Chris Hemsworth</a> says he’s making some lifestyle changes after a genetic test revealed he has a higher-than-average risk of developing <a href="https://www.dailywire.com/topic/alzheimers">Alzheimer’s disease</a>.
The 40-year-old “Thor” <a href="https://www.dailywire.com/topic/entertainment">actor</a> said he’s been changing his diet and workout routines, but has also focused on searching for inner peace. 
“Now, I&#8217;m incorporating more solitude into my life,” Hemsworth told Men’s Health during an <a href="https://www.menshealth.com/health/a45126175/chris-hemsworth-alzheimers-interview/">interview</a> published October 3. “I’ve always been pretty consistent with my exercise commitments, but lately I’ve really felt the importance of taking time for yourself without any outside voice or stimulation and making time for stillness.”
He’s also been incorporating other practices. “I do a lot of meditation and breath work mostly during sauna and ice bath routines,” the Australian actor continued. “For me, my favorite mindfulness work comes from the immersion in physical activities that allow me to be fully present and force me out of me head and into my body, in particular surfing.”
“I have a more consistent approach to my sleep,” Hemsworth continued. “Try to stay off screens an hour before bed and read most nights definitely helps. In addition, focusing on not being attached to every thought and be the observer to the noise when possible, just stepping back from the internal chatter.”
The “Avengers: Endgame” actor has two copies of the gene APOE4, which makes him “eight to 10 times more likely” to develop Alzheimer’s in the future, as The Daily Wire previously <a href="https://www.dailywire.com/news/chris-hemsworth-confronts-mortality-after-discovering-genetic-predisposition-for-alzheimers-disease">reported</a>. He’s part of an estimated 2-3% of the population carrying a copy of the gene from both parents. Alzheimer’s is a progressive type of dementia that affects memory, thinking, and behavior.
Hemsworth said the diagnosis has made him think about his current life more deeply and inspired him to do more “outdoorsy and adventurous” things with his wife and three kids. 
“Doing an episode on death and facing your own mortality made me go, ‘Oh God, I’m not ready to go yet.’ I want to sit and be in this space with a greater sense of stillness and gratitude,” Hemsworth <a href="https://www.dailywire.com/news/from-thor-to-dad-chris-hemsworth-keeps-putting-family-first">said</a> after finding out the news.

Longtime game show host <a href="https://www.dailywire.com/news/bob-barker-beloved-host-of-the-price-is-right-dead-at-99" target="_blank" rel="noopener">Bob Barker</a> — who helmed CBS&#8217; &#8220;The Price Is Right&#8221; from 1972-2007 — died after a battle with Alzheimer&#8217;s Disease.
Entertainment site TMZ was first to <a href="https://www.tmz.com/2023/09/05/bob-barker-cause-of-death-alzheimers-disease-price-is-right/" target="_blank" rel="noopener">report</a> the news, citing the game show icon&#8217;s death certificate. The documents reviewed by the outlet stated that Barker&#8217;s battle with the progressive disease had lasted years — although they did not give specifics regarding when he was diagnosed.
Barker did previously speak out about the disease, teaming up with animal rights group PETA in 2013. Barker and PETA called on especially older people to adopt a vegetarian diet, saying that removing meat from the diet can help to prevent the onset of Alzheimer&#8217;s.

A new study published Wednesday shows that espresso could help prevent Alzheimer’s disease.
Researchers at the University of Verona in Italy found that compounds in espresso beverages, including caffeine, could slow or prevent the brain disorder.
The researchers <a href="https://pubs.acs.org/doi/full/10.1021/acs.jafc.3c01072">concluded</a>, “We have presented a large body of evidence that espresso coffee, a widely consumed beverage, is a source of natural compounds showing beneficial properties in ameliorating … pathologies,” such as Alzheimer’s disease.
The researchers <a href="https://nypost.com/2023/07/19/espresso-might-prevent-alzheimers-development-new-study/">said</a> that “numerous studies” showing that “coffee consumption exerts a [protective] action against two of the most common neurodegenerative diseases, i.e., Parkinson′s and Alzheimer′s,” prompted them to perform the study on espresso because of its high concentration of coffee molecules.
While experts are still uncovering the processes that cause the diseases to develop, most researchers have pointed to a protein in the brain called tau, which helps stabilize structures in that area of the human body. 
However, when tau clumps in the brain, it interrupts the communication between nerves, causing memory loss, poor judgment, wandering, and personality changes — all symptoms of Alzheimer’s disease.
The study used in-vitro and cell culture tests and found that four compounds found in espresso — caffeine, trigonelline, genistein, and theobromine — reduced the length of the tau fibers, preventing them from clumping. As the concentration of caffeine increased, tau proteins could not form larger sheets, indicating that the progress toward Alzheimer’s might be slowed.
Although more research is needed, the researchers said their preliminary findings would “pave the way toward finding or designing other bioactive compounds against … Alzheimer’s,” according to a <a href="https://www.eurekalert.org/news-releases/995534?">press release</a>.
6.2 million Americans <a href="https://www.aplaceformom.com/caregiver-resources/articles/alzheimers-deaths#">have</a> Alzheimer’s disease, and the disorder is the fifth leading cause of death among the elderly, claiming 134,242 American lives in 2020. 
The study was funded by the Italian Ministry of University and Research and follows several studies aimed at linking coffee consumption with reduced risk of Alzheimer’s and Parkinson’s diseases.
A 2018 study by Canadian researchers <a href="https://www.usatoday.com/story/news/nation-now/2018/11/07/dark-roast-coffee-may-reduce-risk-alzheimers-parkinsons-diseases/1917413002/">showed</a> that dark roast coffee is also beneficial for brain health and could work to prevent both Alzheimer’s and Parkinson’s.

However, Howard Bauchner, editor-in-chief of the JAMA peer-reviewed medical magazine, <a href="https://www.usatoday.com/story/news/nation-now/2018/11/07/dark-roast-coffee-may-reduce-risk-alzheimers-parkinsons-diseases/1917413002/">told</a> USA Today at the time that coffee studies should be “taken with a sip of skepticism.” He said that most coffee studies are association studies, meaning they don’t necessarily prove that coffee is the reason for the finding. 
Recent advances in drug therapies have given Alzheimer’s patients and their families hope. A new medication, donanemab, reduced the risk of progressing from mild to moderate dementia by 40% in a recent study and is expected to be <a href="https://www.cnbc.com/2023/07/17/alzheimers-eli-lilly-expects-fda-decision-on-donanemab-by-year-end.html">approved</a> by the FDA this fall.
If approved, donanemab would <a href="https://www.alz.org/media/Documents/alzheimers-dementia-fda-approved-treatments-for-alzheimers-ts.pdf">join</a> Lequembi and Aduhelm as the third FDA-approved drug treatment for Alzheimer’s disease.
Dr. Gil Rabinovici, the director of the University of California San Francisco’s Memory and Aging Center, <a href="https://www.eurekalert.org/news-releases/995699">praised</a> the advancements, saying they were “just the opening chapter in a new era of molecular therapies for Alzheimer’s disease.”

The family of beloved figure <a href="https://www.dailywire.com/topic/celebrities" target="_blank" rel="noopener">“Jungle Jack” Hanna</a> shared that the former zookeeper has been diagnosed with “advanced” <a href="https://www.dailywire.com/topic/alzheimers" target="_blank" rel="noopener">Alzheimer’s disease</a> and can’t remember some of his own children.
Hanna, who resigned as director emeritus of <a href="https://www.dailywire.com/topic/ohio" target="_blank" rel="noopener">Ohio</a>’s Columbus Zoo and Aquarium in December 2020, has dealt with the disease since 2019. He’s credited with building the Columbus Zoo and Aquarium into one of the nation&#8217;s preeminent zoo destinations. He also gained national attention on David Letterman&#8217;s late shows, &#8220;Good Morning America,&#8221; and his Emmy-winning animal show series that is still broadcast on television.
“The Jack people knew isn’t here anymore, but pieces of my husband are. And I’m going to hang onto them for as long as I can,” his wife of 54 years, Suzi Hanna, told <a href="https://www.dispatch.com/story/news/nation-world/2023/06/21/jack-hanna-illness-sick-alzheimers-columbus-zoo-david-letterman-animal-guy-good-morning-america/70293207007/" target="_blank" rel="noopener">The Columbus Dispatch</a>. 
The couple shares three daughters, Kathaleen, Suzanne, and Julie. They say Hanna only remembers his wife and their dog, Brassy, plus sometimes can recognize his eldest daughter Kathaleen.
“He just stopped remembering who I was in all ways. Whether it was in person or by phone, he had no idea I was his daughter,” Suzanne told the outlet. “I think it’s because he didn’t see me as much because I got married so young and I moved away.”
“My husband is still in there somewhere,” Suzanne said of his condition. “There are still those sweet, tender moments — you know, pieces of him that made me and the rest of the world fall in love with him.”
Hanna’s wife said that taking care of Hanna is “real hard some days, but he took care of me all those years, and so it’s my turn to take care of him.”
One anchor of routine in Hanna’s life is his daily two-mile walks with Suzanna. “I want to hold on to these walks as long as I can,” she told the outlet. “I remember the day this all officially started. The day the doctor told us what it was. I’ve just tried to hang on to the little pieces of Jack since then.”
The zookeeper’s family members also told The Columbus Dispatch why they decided to reveal details of their experience with Hanna’s declining health.

“He only retired due to the Alzheimer’s,” she said. “He was embarrassed by it. He lived in fear the public would find out.”
Kathaleen is convinced her father “would have worked until the day he died” if he could have.
“I just want it to be your dad and I for as long as I can,” Suzanne said, addressing her daughters. “The river, the sun, Brassy, our walks. That’s what we have left.”

A group of scientists at Cardiff University have revealed <a href="https://www.dailymail.co.uk/health/article-11958713/New-hope-breakthrough-Alzheimers-treatment-20-years-scientists-uncover-92-genes.html" target="_blank" rel="noopener">stunning information</a> that could lead to life-changing treatment for sufferers of the most common form of Alzheimer’s disease in the next 15 years.
The research by the scientific team started in 2009, and in 2022 they announced they had found three genes that led to the disease. But now they have announced discovering 92 more genes.
“By 2040 I think we&#8217;ll be in the position to offer a range of treatments and we might not know exactly why, but one of them will be able to act on the huge range of causes,” Professor Julie Williams, the Cardiff Dementia Centre&#8217;s director, stated.
“Once you know where to start looking then you can study the effects which genes have on specific brain activity,” Williams said. “Things are speeding up and improving all the time. I&#8217;ve learnt more in the last seven years than I did in the previous 20. Tests which cost millions in the 90s can be carried out for around £30.”
&#8220;For example,” she told the <a href="https://www.bbc.com/news/uk-wales-65166225" target="_blank" rel="noopener">BBC</a>, “we now know that defective genes changing the way immune cells called microglia work. These are the bin lorries of the brain clearing away what they see as rubbish. They may be less efficient at clearing genuine rubbish and mistakenly kill off healthy brain cells, including synapses. Of course synapses are the connections between neurons, so if they get eliminated when they shouldn&#8217;t then you lose connections, you lose thought, you lose memories.”
Williams acknowledged that various therapies would be needed to combat the constellation of problems the disease represents.
Last year, Professor Tara Spires-Jones of the University of Edinburgh said that within ten years, a drug may be available that would limit the suffering of Alzheimer’s patients to initial mild confusion, asserting, “I am wary of using the word cure, which is a very strong word, but I think we will have a disease-modifying drug within 10 years. That is a drug which can stop Alzheimer&#8217;s disease in its tracks, or even &#8211; although this is less likely &#8211; reverse it once it has started.”
“I know all these decades of research can seem disheartening, when there is still no drug for Alzheimer&#8217;s, but game-changing, miracle drugs have been found for other brain diseases, and one is coming for this one,” she added.

Last July, a new computer program was announced that could enable researchers to <a href="https://www.sciencedaily.com/releases/2022/07/220711163210.htm" target="_blank" rel="noopener">diagnose Alzheimer&#8217;s disease much earlier</a> so that it can be treated much earlier after its onset.
Boston University researchers, using audio recordings from neuropsychological interviews with 1,000 people, used speech recognition tools that enabled computers to transcribe the recordings and then translate them into numbers. The chances of cognitive impairment were then determined by those numbers, demographic data, and diagnoses from neurologists and neuropsychologists.
“This approach brings us one step closer to early intervention,” said Ioannis Paschalidis, a co-author on the paper. “It can form the basis of an online tool that could reach everyone and could increase the number of people who get screened early.”

The singer previously described the song as a love letter to his wife, detailing one of the couple&#8217;s happiest moments together in Hawaii.
&#8220;It is dedicated to everyone going through tough times on the journey of life, with the person they care for the most,&#8221; Lydon shared. &#8220;It&#8217;s also a message of hope that ultimately love conquers all.&#8221;
The rocker walked away from the stage several years ago to care for his wife, and had also became an advocate for Alzheimer&#8217;s disease awareness, the outlet noted.
He and the band had also entered the song “Hawaii” in the singing competition Eurovision in hopes of being able to represent Ireland. While he and the band ended up losing the bid to perform the song in the competition, the group is expected to compete at the Eurovision Song Contest in Liverpool next month, the outlet noted.

The cause of death of legendary sex symbol Raquel Welch has been revealed.
Welch, born Jo-Raquel Tejada in Chicago on September 5, 1940, died on February 15 of cardiac arrest after battling Alzheimer’s disease, her death certificate attests, according to <a href="https://www.tmz.com/2023/04/04/raquel-welch-cause-death-certificate/" target="_blank" rel="noopener">TMZ</a>. Her battle with Alzheimer’s had not been revealed at the time of her death.
Welch <a href="https://archive.org/details/FOXNEWSW_20140102_090000_The_OReilly_Factor/start/277/end/337?q=raquel+welch" target="_blank" rel="noopener">told</a> then-Fox News host Bill O&#8217;Reilly in 2014 that she “was more on the conservative side,” adding, “I think it was my upbringing, to tell you the truth, Midwestern values. I just was raised in a certain way, ladies behave a certain way, your country is important, honor those people who do fight for our country.”
Welch accompanied famed comedian Bob Hope when he made his famous visits to entertain American soldiers stationed in Vietnam.

&#8220;Thor&#8221; actor <a href="https://dailywire.com/news/natalie-portman-says-thor-star-dumped-his-carnivore-diet-before-kiss-scene-because-im-vegan/" target="_blank" rel="noopener">Chris Hemsworth</a> has been very busy in recent years — but he plans to take fewer roles in the future due to the results of some genetic tests he had done late last year.
Hemsworth, 39, underwent a <a href="https://dailywire.com/news/chris-hemsworth-confronts-mortality-after-discovering-genetic-predisposition-for-alzheimers-disease/" target="_blank" rel="noopener">series of tests</a> for his National Geographic and Disney+ docuseries &#8220;Limitless&#8221; — and learned that because he had inherited two copies of the APOE4 gene (one each from his mother and his father), he was at greater risk of developing Alzheimer&#8217;s disease than most people.
According to a <a href="https://pagesix.com/2023/04/02/chris-hemsworth-taking-acting-break-after-alzheimers-risk/" target="_blank" rel="noopener">report</a> published Sunday at entertainment site Page Six, a source close to the Australian actor said that after he wraps his current projects — which include a Hulk Hogan biopic and his next appearance as Thor in the Marvel Cinematic Universe — he plans to slow down and make the most of his time with family while his kids are still young.
Hemsworth himself hinted at such a move in a Vanity Fair <a href="https://www.vanityfair.com/hollywood/2022/11/chris-hemsworth-exclusive-interview-alzheimers-limitless" target="_blank" rel="noopener">interview</a> published last November, just a short time after he had learned the results of the genetic tests.
&#8220;Doing an episode on death and facing your own mortality made me go, &#8216;Oh God, I&#8217;m not ready to go yet.&#8217; I want to sit and be in this space with a greater sense of stillness and gratitude,&#8221; Hemsworth explained at the time. &#8220;And then you start talking about kids and family and going, &#8216;Oh my God, they&#8217;re getting older, they&#8217;re growing up and I keep slapping another movie on top of another movie.&#8217; Before you know it, they&#8217;re 18 and they&#8217;ve moved out of house, and I missed the window.&#8221;
&#8220;It really triggered something in me to want to take some time off. And since we finished the show, I&#8217;ve been completing the things I was already contracted to do. Now when I finish this tour this week, I&#8217;m going home and I&#8217;m going to have a good chunk of time off and just simplify. Be with the kids, be with my wife,&#8221; he said.
Hemsworth made sure to drive home the point that he had not been given a hard diagnosis of the progressive condition — only that he had been told he had a higher-than-normal chance of developing the disease later in life.

A new study has found that continuous <a href="https://www.dailywire.com/topic/marriage" target="_blank" rel="noopener">marriage</a>, particularly through the middle decades of life, lowers the risk of developing dementia, Neuroscience News <a href="https://neurosciencenews.com/continuous-marriage-dementia-22373/" target="_blank" rel="noopener">reported</a>. 
Researchers, predominantly from the Norwegian Institute of Public Health, looked at the marital status of adults aged 44 through 68, and whether those adults developed dementia after the age of 70. The study utilized data from 150,000 Norwegians who granted consent for their health information to be used for research. Results indicated adults who remained married during those years of life saw the lowest occurrence of dementia.
Consistent with those results, the data also showed adults who were unmarried and divorced saw the highest rate of dementia. Vegard Skirbekk at the Norwegian Institute of Public Health (NIPH/FHI), and an author of the study, said, “Being married can have an influence on risk factors for dementia.”
The causes of dementia are still unknown, and researchers have found a number of factors that can influence a person’s risk of developing the disease. Such factors were necessarily accounted for in this study.
At the <a href="https://journals.sagepub.com/doi/10.1177/08982643221131926" target="_blank" rel="noopener">study’s</a> conclusion, authors “estimate that, had all participants been continuously married (and shared the same underlying somatic and mental health plus social characteristics of those who marry), 6% of the dementia cases in our study would not have occurred. This is a considerable reduction and is equivalent to the proportion of dementia cases accounted for by smoking and obesity combined, as reported by the Lancet dementia commission in 2020.”
It was found that having children also lowers the risk of developing dementia. Of the unmarried people analyzed, those who had children saw a 60% decreased risk of dementia.
Asta Håberg, a doctor at St. Olav’s Hospital and professor at NTNU, as well as a researcher at NIPH/FHI, commented on the study&#8217;s findings as they pertained to having children.
“Some people have theorized that if you have children, you stay more cognitively engaged. For example, you have to deal with people and participate in activities that you wouldn’t otherwise have to. This stimulates your brain so that it possibly works better. That way you build up a kind of cognitive reserve,” Håberg said.
“Cognitive reserves” are not physically observable in the brain, however scientists understand that there are many things that can build up reserves, which work to prevent dementia. Håberg notes that education is another factor that builds up cognitive reserves.
The results of the study are a part of the research project REFAWOR (Cognitive reserve work and family), which is funded by the U.S. National Institutes of Health. The project seeks to study the relationship between lifestyle and developing cognitive diseases, like Alzheimer’s and dementia, amidst the pressing reality of <a href="https://www.who.int/news-room/fact-sheets/detail/ageing-and-health#:~:text=By%202050%2C%20the%20world's%20population,2050%20to%20reach%20426%20million." target="_blank" rel="noopener">population aging</a> in the decades to come.
It is still unknown what causes this disease that leads to a decline in memory and cognitive function. However, the results of the study lead scientists down more avenues of investigation in order to better understand dementia and how to prevent it.
Håberg sees this study as an exciting opportunity to further investigate the effects that having children has on dementia. At the same time, Skirbekk recognizes further opportunities in understanding the role genetics play in developing dementia. He says that “certain genes increase the risk of dementia,” but it has been observed that people with those genes may never develop cognitive diseases.
The study does not address the biology behind dementia, Skirbekk readily admits. However, “it shows that being married can have an influence on risk factors. You become more cognitively active, you cope better with adversity and are less subject to stress. The partner represents a security that provides a buffer.”

The U.S. Food and Drug Administration (FDA) <a href="https://www.fda.gov/news-events/press-announcements/fda-grants-accelerated-approval-alzheimers-disease-treatment" target="_blank" rel="noopener">granted</a> accelerated approval for a new drug designed to treat Alzheimer’s on Friday.
The drug is Leqembi, which was <a href="https://www.statnews.com/2023/01/06/fda-approves-alzheimers-drug-leqembi-eisai/" target="_blank" rel="noopener">created</a> by Japanese pharmaceutical company Eisai, which will work with Biogen in order to market and commercialize it. The accelerated approval is based on a small study that showed patients who were given the drug “had significant dose- and time-dependent reduction of amyloid beta plaque,” which is an indicator of Alzheimer’s.
When someone has Alzheimer’s <a href="https://www.nia.nih.gov/health/what-happens-brain-alzheimers-disease" target="_blank" rel="noopener">disease</a>, amounts of the beta-amyloid protein group together to <a href="https://www.alz.org/alzheimers-dementia/what-is-alzheimers/brain_tour_part_2" target="_blank" rel="noopener">create</a> plaques in between neurons, impacting how the cells work.
The drug’s price will be an annual $26,500 for someone who is of average weight. Medicare has not <a href="https://www.nytimes.com/2023/01/06/health/alzheimers-drug-leqembi-lecanemab.html" target="_blank" rel="noopener">said</a> it will cover the treatment yet.
Experts of the disease said that the drug offers modest advantages for people who have minor cognitive impairment or early Alzheimer’s disease.
The approval brings to mind Aduhelm, another Alzheimer&#8217;s treatment that was approved by the FDA in June of 2021. The drug was <a href="https://www.nytimes.com/2022/12/29/health/alzheimers-drug-aduhelm-biogen.html" target="_blank" rel="noopener">created</a> by Biogen and many Alzheimer’s scientists spoke out against it. Due to unknown upsides and potential brain swelling and bleeding risks, large health groups chose not to even provide the drug. The treatment was eventually put aside in the marketplace after Medicare restricted its coverage of it. Eisai was <a href="https://www.fiercepharma.com/marketing/biogen-revamps-eisai-alzheimers-disease-pact-taking-full-control-over-troubled-aduhelm" target="_blank" rel="noopener">working</a> with Biogen on the drug, but bowed out last year.
A congressional investigation put forward in December discovered that the FDA’s procedure for approving Aduhelm was “rife with irregularities.” The agency&#8217;s work with Biogen was questionable. The investigation also found that Biogen put a high cost on the drug.
Democratic Representative Frank Pallone of New Jersey <a href="https://www.nytimes.com/2022/12/29/health/alzheimers-drug-aduhelm-biogen.html" target="_blank" rel="noopener">said</a> in a statement that the findings “[document] the atypical F.D.A. review process and corporate greed that preceded F.D.A.’s controversial decision to grant accelerated approval to Aduhelm.”
The FDA’s own internal inquiry, done before the drug was greenlit, discovered that people at the agency didn’t listen closely enough to opposing opinions from the FDA’s statistical team, which argued that there wasn’t enough proof that the drug was successful. It also found that its work with Biogen, the maker of the drug, “exceeded the norm in some respects.”
The new drug, Leqembi, still poses some potential dangers. The FDA noted that it is incorporating a warning for Leqembi over amyloid-related imaging abnormalities (ARIA), which can lead to life-threatening and serious situations, but typically no symptoms are present.
“ARIA most commonly presents as temporary swelling in areas of the brain that usually resolves over time and may be accompanied by small spots of bleeding in or on the surface of the brain, though some people may have symptoms such as headache, confusion, dizziness, vision changes, nausea and seizure,” it noted.
It said the most frequent side effects were headache, ARIA, and “infusion-related reactions.”
 The FDA will also contemplate final approval after it receives more information from a larger study by the company in a few days, which is something it didn’t do with Adehulm, which was also approved in the accelerated format.
The larger, 18-month study of Leqembi <a href="https://endpts.com/fda-approves-alzheimers-drug-lecanemab-from-eisai/" target="_blank" rel="noopener">revealed</a> that 17.3% of patients using the treatment experienced indications of brain bleeding, while 9% of the placebo group had it. Signs of swelling of the brain also <a href="https://www.wsj.com/articles/new-alzheimers-drug-approved-by-fda-promises-to-slow-disease-11673032312" target="_blank" rel="noopener">happened</a> in 12.6% of those who took Leqembi, compared with 1.7% of those who were on the placebo. &#8220;[S]erious adverse events&#8221; <a href="https://www.nytimes.com/2023/01/06/health/alzheimers-drug-leqembi-lecanemab.html" target="_blank" rel="noopener">happened</a> in 14% of those taking Leqembi and 11% of those who got the placebo. Almost 7% of people on Leqembi ended the trial due to adverse side effects, which was more than double the percentage of people who quit who were taking the placebo.
Physicians also <a href="https://www.wsj.com/articles/new-alzheimers-drug-approved-by-fda-promises-to-slow-disease-11673032312" target="_blank" rel="noopener">reportedly</a> said that side effects can be managed with appropriate watchfulness and most patients don&#8217;t have unpleasant symptoms. If scans do find swelling or bleeding, the patient can temporarily stop receiving the drug.
But there is also concern for people who are taking blood thinners. Three people who took the medicine <a href="https://www.inquirer.com/health/fda-approves-alzheimers-drug-leqembi-safety-risks-20230106.html" target="_blank" rel="noopener">passed</a> away while on it, but Eisai said the fatalities can’t be blamed on the medicine. Two of the deceased were taking blood thinners.
The company’s clinical trial also proposed that Leqembi won’t work for around 10% of people who have two copies of APOE4, and they could be hurt by the drug. <a href="https://www.nia.nih.gov/news/study-reveals-how-apoe4-gene-may-increase-risk-dementia#:~:text=APOE4%20is%20the%20strongest%20risk,lipid%20imbalances%20in%20brain%20cells." target="_blank" rel="noopener">APOE4</a> is the most frequent genetic risk factor for the disease. Around 25% of people have one copy of it, and 2% to 3% of people have two copies.
The <a href="https://apnews.com/article/science-health-business-986f1d9a46a48a50a6b554a6c2950801">study</a> showed that people who took the drug were 31% less likely to continue to the next stage of Alzheimer’s while in the study. One kind of swelling was experienced in around 13% of people who got the drug. The treatment also put off the patients’ deterioration by around five months during the study, although the measurement of the decline was not as much as some would have preferred.
“It is unlikely that the small difference reported in this trial will be noticeable by individual patients,” Dr. Madhav Thambisetty of the National Institute on Aging, said in November. He clarified he wasn’t speaking on behalf of the agency.
Some doctors said they will prescribe the drug for patients for whom it might be beneficial, but others caution that it might not do a lot. Physicians also <a href="https://www.wsj.com/articles/new-alzheimers-drug-approved-by-fda-promises-to-slow-disease-11673032312" target="_blank" rel="noopener">note</a> that its impact isn&#8217;t strong and it isn&#8217;t the same as a cure.
“Most patients won’t notice the difference,” Dr. Matthew Schrag, a neurology researcher at Vanderbilt University, <a href="https://www.inquirer.com/health/fda-approves-alzheimers-drug-leqembi-safety-risks-20230106.html" target="_blank" rel="noopener">said</a>. “This is really quite a small effect and probably below the threshold of what we’d call clinically significant.”
Schrag said there is “less drama” for Leqembi when compared with Aduhelm, but he noted that a lot of the similar worries are still there.
“Is this slight, measurable benefit worth the hefty price tag and the side effects patients may experience?” he asked. “I have pretty serious doubts.”
